Acting in Unison
Oh my God, I love it when they talk at the same time!
After so many years together it's not surprising that Dean and Sam will often speak or sing or act in unison. It's really adorable. Below is a list of episodes where this happens with the time of the event included (in minutes and seconds – all times are approximate). Instances where outside forces cause the boys to act in unison (i.e. being thrown against the wall by a demon) are not included.

The boys shoot at the shtriga simultaneously. (36:23)
In response to John's statement that they will work together: “Yes, sir.” (41:10)
Can the boys tell the guest stars of the week the truth: “Nah.” (26:18)
In response to Ellen's question if they are the Winchesters: “Yeah.” (10:02)
The boys show Ronald Resnick their FBI badges at 4:30.
When Bobby asks if they are exaggerating about dancing aliens, they answer: "No." (18:35)
The boys play "Rock, Paper, Scissors" at 11:29 and 11:37.
Deacon tries to get the boys' attention and they respond: “What?” (31:44)

When Cas disappears, the boys shout in frustration: “Cas!?!” at 15:12. Later when they are arguing and Meg keeps interrupting, they tell her “Shut up Meg”, twice, at 23:35 and 23:58.
To put Kevin's fears to rest regarding Crowley, the boys tell him: “OK, he is not in your head.” (2:46)
The boys are brainstorming to find a way to help Charlie and the solution is: “African Dream Root.” (28:52)
After being thanked by Dash for saving everyone, Sam and Dean both enter the Impala at the same time.
Dean and Sam tell each other what the monster is at approximately 41 minutes: "Grigori".
Sam and Dean cock their pistols at the same time after exiting the Impala at approximately 8 minutes.
The Boys respond "No" to Rowena's question about being a team at approximately 40 minutes. Then, at approximately 43 minutes, when Rowena suggests the three of them have a "wee sing-song," Sam and Dean both reply with a "No" again.
Sam and Dean sing along to Bob Seger's "Night Moves."
Sam and Dean flash their FBI badges and then put them back in their breast pockets at approximately 23 minutes.
The Boys play Rock, Paper, Scissors at approximately 38 minutes.

When Crowley offers to help, a skeptical Dean and Sam respond “If...” because they want to know what he wants in return at 10:15.
Sam, Dean and Mary put away their FBI badges at the same time.
When Crowley tells the Winchesters he is "persona non grata" in his own palace. Sam and Dean both respond incredulously with "Palace?"
On occasion, the Winchesters will speak in unison with another character.
“old” Dean and Bobby say “Shut up Sam!” (13:55)
Sam and Lucifer explain to Dean: “You can't torture someone who has nothing left for you to take away.” (9:33)
After Sam announces the victims were killed by belladonna, Dean and Charlie respond: “The porn star?” at 19:34 and upon clarification that Sam means the poison, Dean and Charlie say "oh" at 19:40.
After Sam is surprised that "Carry On Wayward Son" will be sung in the play, Dean and Marie both tell him that it is a "classic." (7:41)
When Rowena wants to know if Sam and Castiel plan on letting her in on what's going on, both respond with a "No."
Castiel and Crowley, posing as FBI agents, both greet Sam and Dean with an "Agent," much to Dean's annoyance.
