Like an archangel, Jack's grace is able to recharge over time. However, while archangel grace can take anywhere from a month to a century to recharge, Jack's half-human nature means his grace will probably lean more towards the longer end of the scale rather than the shorter end.
Without his powers, Jack is as vulnerable and mortal as a regular human. Due to Jack's reliance on his powers for most of his short life, he at first lacks the skills to hunt and fight like a regular human. After saving Lora without his powers, Jack received proper training in hunting from the Winchesters and Castiel. By the time Jack joined the Winchesters, Castiel, and Rowena in a gorgon hunt months after losing his powers, Jack had gained the skills he needed to hunt successfully without special powers.
As the unnatural union of a human and an archangel, his grace was the element that held his being in balance. After Lucifer steals his grace, his cells begin fighting each other, severely weakening Jack and causing his body to slowly shut down until his eventual death. It is only with Enochian magic powered by a piece of Jack's own soul that Jack's condition was able to be cured and his body sustained in his graceless state. However, this cure did not appear to be permanent and Jack appeared to occasionally need to use the magic to heal his illness when it would return. When using this power, a white light would emit from his pupil. By tapping into the power of his soul using the magic keeping him alive, Jack was able to use several of his powers, albeit at the cost of pieces of his soul. After killing the Apocalypse World Michael and absorbing his grace, Jack's original Nephilim powers were restored, eliminating the need for him to further tap into the power of his soul.
After death, Jack's soul is able to go to Heaven like a regular human, though his half-angel nature allows him to sense the true nature of his Heaven and leave it. As he is half-angel, the Cosmic Entity believed that Jack's soul belonged in the Empty upon death like all other angels and demons. With his human soul having been burned away to kill Michael, Jack ends up in the Empty following his second death due to his angelic nature.
