File:Mouth of purgatory.jpg
The portal to Purgatory is opened, moments before Eve rises from its depths.

Dean: Purgatory? Purgatory's real?
Alpha Vampire: Stupid cattle... Of course! And it is filled with the soul of every hungry thing like me that ever walked this earth. Now, where is it? That is the mystery, and that is what your kind-hearted granddaddy is trying to beat out of me.

Dean Winchester, Alpha Vampire, 6.07 Family Matters

It's like the backside of your worst nightmares. It's all blood and bone and darkness. Filled with the bodies and souls of all things hungry, sharp, and nasty.

Bobby, 6.12 Like A Virgin

Characteristics

Purgatory is a spiritual realm where souls of supernatural creatures go after they die. Its physical location is separate from both Heaven and Hell. Although Crowley reports that it is adjacent to Hell[1]. Its entrance is hidden away and blocked by boulders, and its interior seems to be made up of lava[2]. Dragons, after reciting a spell from a book, open a doorway to Purgatory, which releases Eve from its bowels.[2]

6.07 Family Matters

After resurrecting Samuel Campbell, Crowley has him tracking down high-ranking creatures, especially Alphas, so that he can question them about the location of Purgatory. Crowley says he is interested in Purgatory for "development purposes"

Location, location, location. I'm a developer. Purgatory is vast, underutilized, and hell adjacent, and I want it.

6.10 Caged Heat

Crowley tortures and interrogates the Alpha Shapeshifter for Purgatory's exact coordinates. But the Alpha refuses to reveal its address to which Crowley angrily retaliates by decapitating it with a blade, killing it immediately. Crowley's human vessel is destroyed when Castiel incinerates his remains and his essence/spirit may have been sent to Purgatory.

6.11 Appointment in Samarra

Death hints to Dean that he supports their search for Purgatory and encourages him to continue.

6.12 Like A Virgin

Purgatory is revealed not only to be the place where supernatural beings go after death, but also to be the place where the "Mother of All" is locked away. A group of Dragons are revealed to be collecting virgins in an attempt to raise the "Mother of All" (Eve) from Purgatory and allow her to walk the Earth once again. Sam and Dean discover an ancient manuscript made of human skin that Bobby ultimately deduces is a manual for accessing Purgatory and raising Eve from it.

Toward the end of the episode, this plan is carried out by the remaining dragons, who take one of their remaining captives and use the knowledge gained from the manuscript to open a door to Purgatory before throwing the young woman into it. Out of the portal, the girl rises again as the vessel of Eve.
