1.20 Dead Man's Blood
:Vampires: Beau, Kate, Jenny and LutherDaniel Elkins attracts the attention of a group of vampires, led by Kate, who follow him home and kill him after a struggle. They take the Colt from him and Kate presents it to her mate and patriarch, Luther, who scolds Kate at first for killing a hunter. The vampires live in an old barn where they capture tourists and feed on them, turning one such victim, Jenny, into one of their own. Their nest is invaded by the Winchesters, where Kate is lured after them, only to be captured and injected with dead man's blood while another vampire is killed. John Winchester uses her as a bargaining chip to force Luther to hand over the Colt, after which the vampires try to kill John. At the same time, Sam and Dean rescue the vampire's captives, killing the vampire guarding them and then go to help their father. Sam and Dean take down two vampires with dead man's blood soaked arrows and presumably later kill them. John is able to kill Luther with the Colt, while only Kate and Jenny escape alive.

:Vampires: Alpha Vampire, Boris, Dean Winchester, Kristen and Robert
In Limestone, Illinois, Sam and Dean investigate the disappearances of several young women. They discover that the most recent victim was obsessed with Twiligh-style vampires, and that she was communicating with someone who claimed to be a vampire online. At the same time, a blood bank van is robbed. Sam and Dean go to a club called the Black Rose, the girl's last destination, and discover vampires trying to "recruit" new members. Sam kills one vampire, but Dean is turned by a vamp named Boris. Sam intervenes, belatedly, and chases the vampire away. The Winchesters return to their hotel room, and Sam calls Samuel. Dean, who is prepared to be killed by Samuel, sneaks away to bid Lisa goodbye. He is nearly overwhelmed by his hunger, but resists and returns to Sam and Samuel. Samuel surprises Dean by revealing a cure in his grandfather's hunting journal. In order for it to work, Dean must consume no human blood, and must acquire the blood of the vampire that turned him, so Dean goes to the nest of the vampires with a syringe of dead man's blood.

At the nest, a "recruiter" named Robert offers Dean blood, which he declines, and reveals that they are not longer allowed to kill people. Dean meets Boris, and tries to stab him with the dead man's blood, but Boris is too quick. Before Boris can kill Dean, all the vampires collapse, Dean included. He has a shared vision of the Alpha Vampire, and perceives that the Alpha wishes to build an army. When he awakes, Dean must fight the vampires in the nest. He eventually kills all the members—except one beheaded by Sam and Samuel—and gets Boris's blood. Samuel prepares the cure and Dean drinks it. He vomits blood and passes out, but re-awakens as a human.

Castiel brings Lenore to Bobby's place so that they can ask her for Eve's location. She is hesitant, but eventually tells them where to find Eve: in Grants Pass, Oregon. She then asks to be killed. With Eve on earth, her nest has abandoned her and returned to feeding on humans, and even she has given in to her bloodlust on one occasion. Sam and Dean try to persuade her that after Eve is destroyed things will improve, but Lenore is doubtful. Castiel places a hand on her head and kills her.

Flashback: Dean, Benny and Castiel defend themselves from three monsters that attack them and then argue about the fact that Castiel's presence is drawing attention to Dean and Benny. Benny and Castiel want Dean to leave Castiel behind, but Dean is determined to get all three of them out as per their deal or die trying. When Dean points out that Benny is a vampire, Benny insists that he only feeds on blood transfusions and that "I feed on blood, not people." However, before they can argue anymore, Castiel senses Leviathans approaching and three are forced to run. The three end up fighting two Leviathans and Dean decapitates one and Benny decapitates the other, saving Castiel much to Dean's shock.

A rouge vampire named Desmond decides to create his own nest and tries to entice Benny, who has moved back into his hometown in Louisiana and is looking after his great-granddaughter Elizabeth secretly, into helping him. However, Benny refuses so Desmond warns him that he will kill someone every night until Benny agrees. Desmond's first victim draws the attention of mentally-unstable hunter Martin Creaser who is following Benny and believes him to be the killer. He calls in Sam and Dean, but Dean believes Benny's story even after finding him after he buries Desmond's second victim. Despite Dean's belief, Martin and Sam go after Benny until Dean tricks Sam into leaving. Dean and Benny go after Desmond after Benny pretends to agree to join him to learn where he is and in the confrontation that follows, Benny kills Desmond. :Vampire: Alpha Vampire
As part of their American operations, the British Men of Letters begin Project V, a plan to wipe out all vampires in the United States. Armed with the Antianti-Vamp Devicevamp device or AVD, the British operatives kill 230 of the 241 vampires in the Midwest Region of the United States with the remaining vampire retreating to a single nest in Wichita, Kansas. However, the deaths of so many vampires draws the attention of the Alpha Vampire who rallies the remaining Midwest vampires to strike back against the British Men of Letters compound.
At the time of the vampire raid, aided by the fact that hunter Pierce Moncrieff is on the Alpha Vampire's payroll, Mr. Ketch attempts to get Dean to join the British operation by taking him to deal with the nest themselves. In the nest, the two men find a single traumatized vampire that had remained behind. Before they kill her, the vampire reveals that the rest of the nest has gone after the British operation.
When the attack begins, Sam is in the British compound BMOL mobile command as Mary Winchester was trying to entice him into joining the operation by showing Sam what they were up to. The vampires kill two British operatives and breach the compound, but Sam and Mary manage to capture the nest leader and kill another vampire. Before being killed by Mary, the vampire reveals that the Alpha Vampire is leading the raid.
Surrounded with no way to get help, Sam, Mary, Mick Davies, Pierce, Alton Morehead and Serena Colman attempt to come up with a plan to defend themselves, settling on using the AVD as a bug bomb in the compound's vents. With the AVD doubtful to harm the Alpha Vampire, Mick produces The the Colt, the one weapon the British Men of Letters possess that could potentially kill the Alpha Vampire. After providing Mick with the ritual to create more bullets for the Colt, Sam leads Mary and Pierce to get the AVD. Sam is delayed killing two invading vampires, leaving Pierce and Mary to get the AVD. Pierce knocks Mary unconscious, destroys the AVD and gives the Alpha Vampire the all-clear.
With Pierce's help, the Alpha Vampire penetrates the control room and kills Alton and Serena. As the Alpha Vampire attempts to threaten Mick into ending the operation, Sam and Mary return and Mary knocks Pierce out while Sam retrieves the Colt and threatens the Alpha Vampire. In the standoff that follows, Mick manages to slip Sam a bullet for the gun while Mary distracts the Alpha Vampire so that Sam can load the bullet. Sam ultimately shoots the Alpha Vampire through the head with the Colt, killing him.

A group of masked vampires kill a man, Lyle, cut out his wife's tongue and kidnap their children. Investigating, Sam and Dean recognize the wife's drawing of the masks from John's Journal which documents his unsuccessful attempt to hunt the vampires in 1986. Using the pattern documented by John in his journal, Sam and Dean are able to intercept two vampires at their next attack spot, killing one and capturing the other. The vampire admits that his nest kidnaps, raises them up right and then "juices" them because they don't snack on "fast food." In return for a quick death, the vampire directs the Winchesters to the nest.

At the nest, the Winchesters rescue the children, but face off against the vampires who overpower them. One of the vampires is revealed to be Jenny, the young woman that Luther and Kate had turned into a vampire during Sam and Dean's first vampire hunt fourteen years before. As Jenny prepares to feed on Dean, a recovered Sam kills her from behind and fight breaks out. The Winchesters manage to kill the vampires, but Dean is impaled upon a metal spike by one and dies as a result.
