A vampire is created when a person ingests vampire blood, but the transformation is not complete until they feed on human blood.<ref name="one20">[[1.20 Dead Man's Blood]]</ref><ref name="six05">[[6.05 Live Free or Twihard]]</ref> Once turned, they suffer from [[Bloodlust (Pad of Definitions)|bloodlust]] and must feed on blood, human or animal, to survive.<ref name="two03">[[2.03 Bloodlust]]</ref> Unlike folkloric vampires, they do not require an invitation to enter a home and are not killed by sunlight, however prolonged exposure to sunlight can be painful. They take [[the Colt]] from him and Kate presents it to her mate and patriarch, [[Luther]], who scolds Kate at first for killing a [[hunter]]. The vampires live in an old barn where they capture tourists and feed on them, turning one such victim, [[Jenny (Dead Man's BloodVampire)|Jenny]], into one of their own. Their nest is invaded by the Winchesters, where Kate is lured after them, only to be captured and injected with [[dead man's blood]] while another vampire is killed. [[John Winchester]] uses her as a bargaining chip to force Luther to hand over the Colt, after which the vampires try to kill John. At the same time, [[Sam]] and [[Dean]] rescue the vampire's captives, killing the vampire guarding them and then go to help their father. Sam and Dean take down two vampires with dead man's blood soaked arrows and presumably later kill them. John is able to kill Luther with the Colt, while only Kate and Jenny escape alive.

While trying to learn about the circumstance that lead to the death of his fiancé, [[Ennis RothRoss]] sneaks into the monster club in the back of the restaurant he was dining the night prior. He quickly comes face to face with an employee of the restaurant, who reveals himself to be a vampire. Ennis fires his weapon into the vamp, which has no effect, just as it is about to feed on Ennis, Sam and Dean arrive cut off its head.

A group of vampires who consume all of their prey, not just the blood, go to Hibbing, Minnesota in order to find [[Sheriff Len Cuse]], their former leader with the hope of getting him to rejoin them. Len had left the nest as he got tired of the cries of their prey and started feeding off of blood bags and protecting humans as a sheriff. They kill and consume two people, stealing one's belt and the other's wallet. Despite not being part of the nest anymore, Len does his best to protect them, but the kills draw the attention of [[Sheriff Jody Mills]] who is at his sheriff's retreat. [[File:Starr'sNest.jpg|thumb|right|350px|[[Starr]]'s nest hold their next victims captive.]] She calls the Winchesters to let them know and investigates the kills. After Len is seen over a victim by [[Sheriff Donna Hanscum]], the Winchesters and the sheriffs track the vampires to an old farmhouse where they are captured. [[Starr]] explains the nest's way of operating and tries to get Len to rejoin them by killing the Winchesters and the two sheriffs. However, Len refuses so Star kills him. Dean breaks free and kills Catfish and the other vampire while Star goes after Jody for knocking her down. However, Donna kills her to save Jody.

As part of their American operations, the [[British Men of Letters]] begin [[Project V]], a plan to wipe out all vampires in the United States. Armed with the [[Antianti-Vamp Devicevamp device]] or AVD, the British operatives kill 230 of the 241 vampires in the Midwest Region of the United States with the remaining vampire retreating to a single nest in Wichita, Kansas. However, the deaths of so many vampires draws the attention of the [[Alpha Vampire]] who rallies the remaining Midwest vampires to strike back against the [[British Men of Letters compound]].

When the attack begins, Sam is in the British compound BMOL mobile command as [[Mary Winchester]] was trying to entice him into joining the operation by showing Sam what they were up to. The vampires kill two British operatives and breach the compound, but Sam and Mary manage to capture the nest leader and kill another vampire. Before being killed by Mary, the vampire reveals that the Alpha Vampire is leading the raid.

Surrounded with no way to get help, Sam, Mary, [[Mick Davies]], Pierce, [[Alton Morehead]] and [[Serena Colman]] attempt to come up with a plan to defend themselves, settling on using the AVD as a bug bomb in the compound's vents. With the AVD doubtful to harm the Alpha Vampire, Mick produces [[The the Colt]], the one weapon the British Men of Letters possess that could potentially kill the Alpha Vampire. After providing Mick with the ritual to create more bullets for the Colt, Sam leads Mary and Pierce to get the AVD. Sam is delayed killing two invading vampires, leaving Pierce and Mary to get the AVD. Pierce knocks Mary unconscious, destroys the AVD and gives the Alpha Vampire the all-clear.
