The vampires live in an old barn where they capture tourists and feed on them, turning one such victim, [[Jenny (Dead Man's BloodVampire)|Jenny]], into one of their own. Their nest is invaded by the Winchesters, where Kate is lured after them, only to be captured and injected with [[dead man's blood]] while another vampire is killed. [[John Winchester]] uses her as a bargaining chip to force Luther to hand over the Colt, after which the vampires try to kill John. At the same time, [[Sam]] and [[Dean]] rescue the vampire's captives, killing the vampire guarding them and then go to help their father. In Limestone, Illinois, [[Sam]] and [[Dean]] investigate the disappearances of several young women. They discover that the most recent victim was obsessed with ''Twiligh''-style vampires, and that she was communicating with someone who claimed to be a vampire online. At the same time, a blood bank van is robbed. Sam and Dean go to a club called the Black Rose, the girl's last destination, and discover vampires trying to "recruit" new members. Sam kills one vampire, but Dean is turned by a vamp named [[Boris]]. Sam intervenes, belatedly, and chases the vampire away. The Winchesters return to their hotel room, and Sam calls [[Samuel Campbell|Samuel]]. Dean, who is prepared to be killed by Samuel, sneaks away to bid [[Lisa]] goodbye. He is nearly overwhelmed by his hunger, but resists and returns to Sam and Samuel. Samuel surprises Dean by revealing a cure in his grandfather's hunting journal. In order for it to work, Dean must consume no human blood, and must acquire the blood of the vampire that turned him, so Dean goes to the nest of the vampires with a syringe of [[dead man's blood]].

At the nest, a "recruiter" named [[Robert]] offers [[Dean]] blood, which he declines, and reveals that they are not longer allowed to kill people. Dean meets Boris, and tries to stab him with the dead man's blood, but Boris is too quick. Before Boris can kill Dean, all the vampires collapse, Dean included. He has a shared vision of the [[Alpha Vampire]], and perceives that the [[Alpha]] wishes to build an army. When he awakes, Dean must fight the vampires in the nest. He eventually kills all the members—except one beheaded by Sam and Samuel—and gets Boris's blood. Samuel prepares the cure and Dean drinks it. He vomits blood and passes out, but re-awakens as a human.
Afterwards, the incapacitated vampire is killed and as Annie never fed, Sam and Dean are able to use Celia's blood to create the cure and save her though the cure will make her sick for a few days.

A group of vampires who consume all of their prey, not just the blood, go to Hibbing, Minnesota in order to find [[Sheriff Len Cuse]], their former leader with the hope of getting him to rejoin them. Len had left the nest as he got tired of the cries of their prey and started feeding off of blood bags and protecting humans as a sheriff. They kill and consume two people, stealing one's belt and the other's wallet. Despite not being part of the nest anymore, Len does his best to protect them, but the kills draw the attention of [[Sheriff Jody Mills]] who is at his sheriff's retreat. [[File:Starr'sNest.jpg|thumb|right|350px|[[Starr]]'s nest hold their next victims captive.]] She calls the Winchesters to let them know and investigates the kills. After Len is seen over a victim by [[Sheriff Donna Hanscum]], the Winchesters and the sheriffs track the vampires to an old farmhouse where they are captured. [[Starr]] explains the nest's way of operating and tries to get Len to rejoin them by killing the Winchesters and the two sheriffs. However, Len refuses so Star kills him. Dean breaks free and kills Catfish and the other vampire while Star goes after Jody for knocking her down. However, Donna kills her to save Jody.

After [[Maggie (Apocalypse World)|Maggie]] is captured by the [[djinn]], [[Neil (Djinn)|Neil]], he uses his enhanced powers to manifest Maggie's greatest fear -- a vampire from [[Apocalypse World]] which were responsible for the death of her family. When [[Sasha Rawling]] hears a strange noise coming from the attic, she goes to investigate, but is thwarted by the appearance of the vampire, who scares her off, but does not give chase. Finding Maggie strung up in the attic, having her blood drained, she warns Sam of the vampire, which attempts to attack Sam, but is pierced in the chest and explodes into dust.

A group of masked vampires kill a man, Lyle, cut out his wife's tongue and kidnap their children. Investigating, Sam and Dean recognize the wife's drawing of the masks from [[John's Journal]] which documents his unsuccessful attempt to hunt the vampires in 1986. Using the pattern documented by John in his journal, Sam and Dean are able to intercept two vampires at their next attack spot, killing one and capturing the other. The vampire admits that his nest kidnaps, raises them up right and then "juices" them because they don't snack on "fast food." In return for a quick death, the vampire directs the Winchesters to the nest.
 
At the nest, the Winchesters rescue the children, but face off against the vampires who overpower them. One of the vampires is revealed to be [[Jenny (Vampire)|Jenny]], the young woman that [[Luther]] and [[Kate (vampire)|Kate]] had turned into a vampire during Sam and Dean's first vampire hunt fourteen years before. As Jenny prepares to feed on Dean, a recovered Sam kills her from behind and fight breaks out. The Winchesters manage to kill the vampires, but Dean is impaled upon a metal spike by one and dies as a result.
